This review consolidates preclinical and clinical evidence elucidating ketamines neurochemical cascade: blockade of extrasynaptic NMDA receptors on GABAergic interneurons, resultant glutamate surge, AMPA receptor activation, BDNF release, downstream mTOR signaling, and synaptogenesis in the medial prefrontal cortex and hippocampus.
